To ascertain this hypothesis, further investigation is needed to address questions such as: how do mycotoxins induce this disease, what is the molecular mechanism, and does the brain-gut axis play a role in this context? Immune evasion within trichothecenes was further explored in recent studies. Moreover, the function of hypoxia in this process is notable. However, investigating if this evasion capability is present in other mycotoxins, particularly aflatoxins, is crucial. Our primary focus in this work was on key scientific questions concerning the mechanistic underpinnings of mycotoxin toxicity. We dedicated substantial effort to research questions involving key signaling pathways, the equilibrium of immunostimulatory and immunosuppressive effects, and the connection between autophagy and apoptosis. The discussion also touches upon interesting subjects, such as the complex interplay between mycotoxins and aging processes, as well as the cytoskeleton and its role in immunotoxicity. We have compiled for Food and Chemical Toxicology a special issue on “New insight into mycotoxins and bacterial toxins toxicity assessment, molecular mechanism and food safety,” a crucial undertaking. Researchers are highly motivated to submit their current work for publication in this special issue.
Shellfish and fish serve as a rich source of nutrients essential for fetal development, especially docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA). The issue of mercury (Hg) pollution's impact on fish consumption, particularly for pregnant women, could hinder the development of their children. In Shanghai, China, this study sought to evaluate the risk-benefit profile of fish consumption for expectant mothers, culminating in specific recommendations.
Data from a representative sample of the Shanghai Diet and Health Survey (SDHS) (2016-2017) in China were used for a secondary cross-sectional analysis. Based on a food frequency questionnaire (FFQ) focused on fish, and a 24-hour recall, dietary intake of mercury (Hg) and DHA+EPA was estimated. Raw fish samples (59 common types) from local Shanghai markets were procured and analyzed for their content of DHA, EPA, and mercury. The FAO/WHO model determined population-level health risk and benefit by examining net IQ point gains. High-DHA+EPA, low-MeHg fish were categorized, and the consumption frequency (1, 2, or 3 times per week) of these fish, along with IQ scores, was simulated to estimate their impact on 58 IQ points.
A daily average of 6624 grams of fish and shellfish was consumed by pregnant women in Shanghai. The mean concentrations of mercury (Hg) and EPA+DHA in commonly consumed fish species in Shanghai were 0.179 mg/kg and 0.374 g/100g, respectively. A mere 14% of the population surpassed the MeHg reference dose of 0.1g/kgbw/d, contrasting sharply with the 813% who fell short of the recommended daily intake of 250mg EPA+DHA. According to the FAO/WHO model, the maximum attainable IQ point gain was 284%. With exceptional antifungal activity across a broad spectrum, SYP-3343, a novel strobilurin fungicide, nonetheless raises concerns regarding its potential toxicity to public health. Nevertheless, the vascular harm induced by SYP-3343 on zebrafish embryos remains poorly understood. This study explored the impact of SYP-3343 on vascular development and its underlying mechanism. Zebrafish endothelial cell (zEC) migration was inhibited by SYP-3343, which also altered nuclear morphology, triggered abnormal vasculogenesis and zEC sprouting angiogenesis, ultimately causing angiodysplasia. RNA sequencing experiments showed that exposure to SYP-3343 resulted in changes to transcriptional levels related to vascular development processes in zebrafish embryos, such as angiogenesis, sprouting angiogenesis, blood vessel morphogenesis, blood vessel development, and vasculature development. Zebrafish vascular defects induced by SYP-3343 treatment were ameliorated by the inclusion of NAC. In HUVEC cells, SYP-3343's influence manifested as changes in cell cytoskeleton and morphology, alongside the obstruction of migration and viability, the disruption of cell cycle progression, the depolarization of mitochondrial membrane potential, the promotion of apoptosis, and the elevation of reactive oxygen species (ROS). SYP-3343 treatment led to a disruption of oxidation-antioxidant homeostasis and prompted changes in the expression of genes regulating cell cycle and apoptosis within HUVECs. SYP-3343 demonstrates high cytotoxicity, probably through mechanisms involving the upregulation of p53 and caspase3, and modification of the bax/bcl-2 ratio, which are both influenced by reactive oxygen species (ROS). Hypertension is more frequently observed in Black adults than in both White and Hispanic adults. Even so, the reasons for a greater incidence of hypertension among Black people are uncertain, but environmental chemical exposure, specifically volatile organic compounds (VOCs), could play a role.
A subset of the Jackson Heart Study (JHS) consisting of 778 never-smokers and 416 age- and sex-matched current smokers was used to investigate the associations of blood pressure (BP) and hypertension with volatile organic compound (VOC) exposure. Following adjustment for covariates, metabolites of acrolein and crotonaldehyde were found to be associated with elevated systolic blood pressure, specifically by 16 mm Hg (95% CI 0.4, 2.7; p=0.0007) and 0.8 mm Hg (95% CI 0.001, 1.6; p=0.0049) among non-smokers, while a 0.4 mm Hg (95% CI 0.009, 0.8; p=0.002) increase in diastolic blood pressure was associated with the styrene metabolite. The systolic blood pressure of current smokers was found to be 28mm Hg higher, with a 95% confidence interval ranging from 05 to 51. Hypertension risk was substantially elevated (relative risk of 12; 95% confidence interval, 11-14) for this group, which also exhibited elevated urinary levels of several VOC metabolites. Smoking was linked to higher levels of acrolein, 13-butadiene, and crotonaldehyde urinary metabolites, and this was correspondingly associated with higher systolic blood pressure. The association was markedly stronger among male participants who were under 60 years old. In a study employing Bayesian kernel machine regression to analyze the effects of various VOC exposures, we discovered that acrolein and styrene were the primary determinants of hypertension among non-smokers, while crotonaldehyde exerted a similar effect in smokers.
Possible causes of hypertension in Black populations include environmental VOC exposure and tobacco smoke.
